Specialty and Diagnostic Enzymes

Iosynth is best known for supplying enzymes for biocatalysis, however enzymes are also routinely used for multiple other applications. Please check back regularly as this is a continually expanding portfolio of innovative enzymes.

Proteinase K

Proteinase K is a nonspecific serine protease with a broad substrate specificity that can be used in a wide variety of life science applications such as:

  • Isolation of genomic and plasmid DNA
  • Isolation of RNA
  • Removal and inactivation of phosphatases, nucleases and RNases
  • Improve the cloning of PCR products
  • Protein fingerprinting by mass spectrometry
  • Iosynth supplies lyophilized Proteinase K in bulk quantities, up to multi kg levels.

    rTrypsin

Trypsin (EC 3.4.21.4) is an endopeptidase that preferentially hydrolyses ester bonds whose carboxyl groups are contributed by Lysine (Lys) or Arginine (Arg) except when either if followed by proline (PRO). It is used for a wide range of biotechnological processes. Although native trypsin from bovine or porcine pancreas has been available for many years, a recombinant Trypsin manufactured by microbial fermentation has many benefits.

    Advantages of rTrypsin

  • Produced by a recombinant manufacturing process, rTrypsin is not contaminated with animal enzymes such as chymotrypsin or carboxypeptidases
  • Animal origin-free (AOF) manufacturing process eliminates the risk of viral or animal related contaminants in the final product
  • Simple replacement of bovine or porcine trypsin
  • Stability is not affected by Ca2+
  • Consistent supply
  • Applications for rTrypsin

  • Cell dissociation and propagation of multiple cell lines including stem cells
  • Vaccine production and processing
  • Dissociation of siddected cells prior to cell fixing and sorting
  • Cleavage of proinsulin to yield intact insulin
  • Iosynth supplies rTrypsin as a granulated product in bulk. Before use, a filtration process of the rTrypsin in solution is required.

    Beta Lactamase

Beta Lactamases are a critical enzyme for antibiotic sterility testing and environmental monitoring. Eucodis Bioscience, manufacturer of the famous and trusted LacBuster brand of beta lactamase enzymes, is Iosynth’s partner in this segment.

    Sterility Testing

US Pharmacopeia (USP) Chapter 71 and EP Section 2.6 describe sterility testing of bulk antibiotics, which should be shown to be free from microbial contamination. The testing requires the removal of significant amounts of active antibiotic from solution by combined filtration and the use of penicillinase or β-Lactamase, The resulting solution is tested for the (lack of) growth of microbes.

Our beta-lactamase has significantly higher activity against clinically important beta-lactams, it inactivates clinical relevant cephalosporins better than any other product available on the market.

    Environmental Monitoring Plates

Contact plates, settle plates and air monitoring systems for testing of aseptic conditions in antibiotic manufacturing facilities need to be manufactured with agar medium for neutralization of antibiotic. This is achieved by the addition of Penicillinase or β-Lactamase to the medium. Through this, any antibiotic residues are hydrolyzed and microbial contamination can be detected.

Our Beta-Lactam agar plates are available as contact and settle plates for high performance monitoring of your isolators and clean rooms during beta lactam manufacture.
